Photorepair prevents the induction of apoptosis by transcription arresat and P73 after UV irradiation in cho cells (2001)

- Resumo: Objectives: Study the early signals to UV-induced apoptosis in normal and repair deficient cells (mutated in the XPB gene). Methods and Results: We utilize Chinese Hamster Ovary cells (normal and XPB mutated ) expressing a marsupial photolyase that repairs CPDs, generating the cell lines CHOphr and CHO-XPBphr, respectively. These cell lines when UV irradiated and maintained in light conditions present a prevention in the apoptotic program after flow citometry analysis. When the transcriptional rate was accessed, CHOphr cells present a recovery in transcription synthesis after photorepair. The cells CHO-XPBphr show a similar response. The study of participation of PARP in apoptosis by flow citometry and RRS demonstrated that the (ADP)ribosylation process is involved in UV-induced apoptosis probably acting in transcription arrest. In Western blot experiments, we can observe the prevention of the induction of the P73 damage sensor protein after CPD elimination in the cell lines CHOphr and CHO-XPBphr Conclusion: These results indicate that CPDs are the first signal to UV-induced apoptosis in our system, followed by transcription arrest and induction of P73 and PARP. The elimination of CPDs by heterologous photorepair decreases the early signals that commit the cell to death. The XPB mutation may limit the access of the photolyase enzyme to CPD removal and the participation of PARP in apoptosis
