Identification, subgroup and genotype characterization of human rotavirus circulating in Paraguay: detection of mixtures with G5 and G10 animal rotavirus (2001)

- Resumo: Human rotavirus (HRV) is the major etiologic agent of severe diarrhea in infants and young children worldwide. Although detection of rotavirus in clinical specimens has become routine diagnostic procedure, characterization of this virus has remained difficult despite the need for such information before the introduction of a human rotavirus vaccine. Objectives: Detection and characterization of human rotavirus circulating in Paraguay. Methods and Results: In this study we analyzed 166 fecal samples of hospitalized and ambulatory children patients with acute diarrhea syndrome, mostly under 5 years old, collected in Paraguay from January 99 to September 99. The identification of HRV was performed by an enzyme immunoassay (EIARA-FIOCRUZ) and by RNA electropherotyping. Sixty samples (36,1 %) that were found positive for HRV were then submitted to the subgroup characterization by using an enzyme immunoassay with monoclonal antibodies (MAb. Elisa) against group A, and subgroups I and II. Molecular characterization of these samples was performed by using reverse transcription (RT) followed by polymerase chain reaction (PCR) with specific primers for the most common human and animals G and P genotypes. Results were: 43 (71,6 %) group A/subgroupII; 10 (16,6%) groupA/ subgroup I and II; 7 (11,8%) negative. For G genotypes results were: G[4] (33,3 %) followed by G[1](5,4%); several mixtures like G[4] with G[5]; G[1] with G[10] ; G[1]with G[5]; and G[4] with G[5]and G[10], were also found. For the P types, the most frequent was P[8] (50%), followed by a mixture of P[8][1] (10%), P[4] and P[1] without mixture were also found. Conclusions:These results shows for the first time the diversity of HRV strains circulating in Paraguay and should be regards as preliminary.
