- Abstract: The occurrence of several clinical forms of human paracoccidioidomycosis (PCM) as well as the polar forms of experimental murine infection has been attributed to characteristics of the host and also to intrinsic factors of the fungus. Virulence is one of the most important factor associated with fungi. In the present study, genomic DNA from virulent and non-virulent P. brasiliensis isolates were analysed by RAPD technique to investigate the correlation of genetic difference and the virulence of isolates. Our results confirmed the efficiency of RAPD assay to distinguish P. brasiliensis isolates. 12 primers were utilized and all were able to amplify polymorphic fragments. The primer OPG-03 was the one that showed the largest number of polymorphic fragments, although OPN-10 and OPN-06 also showed fragments that could differentiate the isolates. Two groups were determined: group I comprised one non-virulent isolate, two isolates from patient with chronic PCM (which is considered a mild form of the infection) and one virulent isolate; group II comprised seven non-virulent isolates. It was found that genetic differences could not correlate with virulence. Also it was demonstrated that the loss of the virulence was not related to genetic differences, since the virulent Pb18 isolate and six different Pb18 isolates (Pb18-N) which had lost their virulence after many subcultures were found to be genetically indistinguishable, presenting a similarity coefficient of 95%
