Contribution of NTS and VLM catecholamines cells to the innervation of the median preoptic nucleus (MnPO) (2000)

- Resumo: Objetivo: Previous results demonstrated that the nucleus of the tractus solitarius (NTS) and the ventrolateral medulla (VLM) are major afferents of the MnPO. However the chemical nature of the cells originating these projections remain unknown.In this study we sought to determine the relative contribution of NTS and VLM catecholamines cells to the innervation of the MnPO.Métodos e Resultados: Wistar rats (N=4) weighing 150-200g received microinjections of FluoroGold (2%, 50 nL) orfluorescein microbeads (100-150 nL) into the MnPO. After 7 to 10 days, animals were anesthetized and perfused with 4% paraformaldeyde in phosphate buffer. Serial coronal sections (40 ?m) of brainstem were cut and processed for the presence oftirosine hidroxilase (TH) by indirect immunofuorescence.Results obtained demonstrate that: a. within NTS retrogradelly labeled cells were mostly found in the medial and comissural subnucleus; b. 60% of NTS retrogradelly labeled neurons were also TH-positive; c. within VLM, MnPO projectingcells were mainly found in the caudal VLM; d. 70% of the VLM retrogradelly labeled neurons were also TH-positive.Conclusões: Results obtained demonstrated that cells from C1/C2 or A1/A2 groups are major afferents of the MnPO, and may representthe anatomical substrate of the modulatory role of catecholamines in drinking and hydroeletrolytic balance regulation
